The aim of the project is to develop a novel fluorocarbene transfer reagent based on carbonyl or sulfonium salt precursors and explore its application in synthesis of new fluorinated building blocks. This will be achieved by following objectives:

1) Development and synthesis of novel fluorocarbene source – carboxylic acid fluorohalomethyl ester or fluorhalomethyl sulfonium salt as a synthetic equivalent of CHF synthon.

2) Improved synthetic methodology exploiting novel fluorocarbene source to obtain fluorinated small rings, X-H and C-C insertions, fluorocarbene cross-coupling products.

3) Validation of developed methodology on biologically significant drug molecules via introduction of fluorinated bioisosteres.

Expected outcomes: Novel and versatile monofluorocarbene source solving the long standing problem of non-existent fluorodiazomethane chemistry. Advantageous synthetic approach to access otherwise challenging fluorinated scaffolds in synthetic and medicinal chemistry fields.
